It says "The soft cloth backing means they fit directly on to your current Velcro pads".
Reckon they could be attached without removing the current pads with double-sided tape or similar? Something that wouldn't ruin the existing pads would be best.

Is that between standard and 404 or 404 and 808?
That's between a standard 32 spoke wheel and an 808 at 25km/h, front wheel only.
2W in a 25 might get you from 1:00:00 to 59:52, although at that speed an 808 gets you more like 8W, so about 30s.

Velcro
My current pads are attached with velcro. These Fizik ones are cloth backed as they expect you to remove your existing pads and install the Fiziks. I'm thinking about double the fun and using two sets of pads.

2W in a 25 might get you from 1:00:00 to 59:52, although at that speed an 808 gets you more like 8W, so about 30s.
So, assuming I was to achieve my target 740k (ha! Well, I've got nothing else to aim for), using an 808 over a standard front would get me 4-5 extra kilometres. I'll say "it's not worth it" now and then miss some milestone by 1k and then regret using a standard wheel forever..

If you're after more comfort for the 24h, are you sure gel is the way to go? Seems like a lot of money if you're not certain they are going to be an improvement on the über-comfy 3T or Vision pads.

I already have gel pads. I wanted to get thicker version of the same thing though. Pro don't seem to do them.
To be fair, I made a note of changing pads quite early into my long TT rides but my forearms aren't getting nearly as beaten up as they were.
Not sure those Vision pads would fit the Pro armrests that well. Looks like I'll be sticking with what I got.
