It's made me wonder about rolling resistance and how it changes with tyre size (if it does- I'm making an assumption)
All else being equal, wider tyres have less rolling resistance
and whether bigger tyres are simply a better choice (if we ignore frontal area)
They are, but you can't go on ignoring frontal area forever, otherwise Paris-Roubaix would be contested on fatbikes.

I know nothing about thru axles and that kind of stuff, but looking at that frameset, is thru axles (with flat mounts if I understand correctly?) combined with vertical dropouts (with post mounts, again if I understand this? Unless this has no justification for rear brake?) a bit of a weird thing / likely to mess groupset consistency?
Nice bike otherwise, frameset seems well priced... -

I know nothing about through-axles and discs I confess. I mainly liked it because it's Ti and has more road-bike geo with space for reasonable large volume tyres - it's bang in the middle of the two Kinesis bikes I couldn't choose between. And quite a bit cheaper...

Through axle is not necessary on the rear, due to the different angle between the brake reaction and the wheel removal vectors. On the front, through axle is necessary because disc brakes tend to eject the wheel.

CAAD12 has mismatch front and rear standard, flat mount rear, post mount front.
Bingo, 38mm was just about idea for on and off road.
