Geo here: http://www.giant-bicycles.com/en-us/bikes/model/defy.5/9014/48856/
I guess the first number/reference size is the seat tube.
Xs/43.0 | 51.5
s/46.5 | 53
m/50.0 | 54.5*I *doubt that you will need a 54.5cm. 53cm sounds in the right ballpark. The advantage with that is it has a normal seat post so there's some flexibly for you to get an inline or one with more layback if you need it.
Never be tempted to go for a bike that's too big. Especially if you're paying for a new bike.

I also reckon that for the money you could do slightly better. New you're never going to get a great deal more kit, but....
This Jamis is about the same and has 9 speed sora, with a tigra rear and Mavic CXP22 rims... plus some other little bits.

Also you can go to one of the many Evan's and try it out first for size rather than gambling. Assuming it's not a massive ball ache maybe find out which ever one ed works in and go there.
EDIT: also saw the Jamis frame is made by Kinesis - so that's another plus.

@Ollyit, I'm slightly shorter than you at 5'7" and have a Giant TCR in a Medium. The seattube and cranks (172.5mm) suit me fine, but the reach was a little far for my stubby arms and I've swapped the 110mm stem for a 90mm .
All is good!
Incidentally, why are you getting the Defy 5? There's a Defy 4 on sale at Ash Cycles for £439... http://www.ashcycles.com/site/giant-defy-4-triple-2011?zenid=3a482722cfd40b58136c1f9c37ec09ff

I really would be suprised if a medium would fit properly.
Also just realised that the giant doesn't have carbon forks. Either go cheap for that ^ one for £440, or go for the Jamis (or similar) and get carbon forks and the next tier GS up.

^ it's nice to have these things confirmed though. Especially if you are, like me, a chronic self-doubter.

Bear in mind it does always cover everything, some roadie like to be a little longer after doing core strength, tourer tend to want the handlebar higher, etc.
I am the same height as you and have a TCR - you need at least a small - maybe xs.
