I've mentioned elsewhere in this thread that I've got a bike with an old Arione which I can ride all day long and an almost new (second hand off ebay) identical saddle that I get uncomfortable on in a couple of hours of riding. As far as I can tell, both bikes are set up the same.
So, do these saddles wear in over time? It does seem to be more comfortable with every ride.
