I had this last week while working on a conversion, it was just a stiff link but it went away after slackening the chain tension. Try turning the cranks really slowly and wait for the click, see which link is about to go round the sprocket/chainring and mark it. Spin a few more times and repeat. If its the same place then its a stiff link.
