If you had a choice, would you go with 6 bolt or centre lock? It's for a road bike, running shimano group sets. Does it matter?

Well technically speaking there is a bit of play in a 6 bolt rotor until you tighten the bolts so it’s possible to bolt it on not completely straight. CL is dead straight every time

It does if in the future the majority of quality hubs and rotors are only offered in CL flavour.
