The record is 44 hours and a bit. I'm not aware of an unsupported record.
There are loads of route variations but the fastest involves some not very pleasant roads. Also, while LE is not too far from Penzance station, JoG is not an easy place to get away from - unless you ride on and go for 1,000 miles.
