Been a few years since I was there but Curve had some pleasant routes if you don't mind a bit of gravel:

The latter has the 'Boule' on it (Yarra Boulevard) and might go past the flying fox colony. It's a nice ride away from traffic for a chunk of it too.
Other option could be to head over Westgate bridge and down the coast to Altona, if you time it right you can get the punt back into the city. Used to be able to get most of the way there on cycle paths.

depends on where your hotel is but could head from abbotsford convent out past kew all the way towards warrandyte

Not far from Parliament House
I've borrowed a bike for a few days next week... anyone got any routes that for 1-2 hours of riding. Staying in the CBD
