How many sections are you going to do it in? Depends where you are crossing really, are you are doing bridge across to Saltash, Torpoint ferry or Cremyll ferry. Are you riding via Torbay/Ashburton? Personally I would go via Exeter/Okehampton/Lewdown/ Tinhay/Lifton/Launceston on the old A30.
I have an audax route at home that avoids the horrible horrible horrible A38. It's essentially the motorway for Cornwall.

The old A30? I'd stay well away from any of the major A roads in Cornwall (I grew up there). You can plot lovely back road routes most of the way down.
When riding down from London a few years ago I went across Dartmoor and crossed the border at Horsebridge (having lunch in the amazing old pub there - worth calling ahead) before taking the back lanes through Minions before hopping on the A38 for 10mins or so and then heading over to Bodinnick and the ferry over to Fowey. You could then head out along the Roseland coast towards the King Harry ferry at Falmouth and off via the lanes to Lands End.
The coast is far prettier than riding through clay country and it's quieter on the south.

The old A30 is the one that runs parallel to the new A30, and is extremely quiet because it winds through the little villages, and fairly easy riding if you are grinding out some distance. Nice food stop at Cheriton Bishop.
Planning a cycling trip from Essex to Land's end, all routes are done, except the last one.
Does anyone try Plymouth to Land's end before or familiar with the route around there?
Should I ride along the coast via St Austell or go inland?
Cheers
