Yes if it's a cassette on a free hub
Or rearrange the cogs so the gear you want is on the right chainline.If it is a freewheel block then you would need to screw on a single speed freewheel and redish the wheel for the right chainline.

no these will bring your chain line into line by adjusting where you put the spacers
or you could use one of these as there are numerous spacers of various sizes shouild be easy to get the right chainline - onza single speed conversion kit

So I am building a SS bike, I have a frame with 130 spacing & horizontal drop outs. ( all city nature boy )
I have been offered a set of road wheels, that will do the job.
Can I just just spacers on free body to hold the free wheel cog in place ?
