I would think about where you're going to cross A40 Westway... I personally would, if you're heading for Regents park north, head for Royal Oak underpass perhaps, by way of Maida Vale, to avoid Marylebone/Edgware Rd/paddington. Then thread a route through notting hill.
Enjoy it! Long commutes are great when you stop rushing and start enjoying.

I do Hackney Road to Shepherds Bush every day. You need to either stay a little North as suggested, or a little South of town. Don't touch Euston Road.

This is a good way of doing part of the route:

Not back streets but not busy, and the best way I've found of crossing the Westway -

I used to do Walthamstow to Ladbdroke Grove- the N End
It was , I think, ride to Essex Rd - then down to the Bloomsbury Cycle route at Packington street, its a bit hard to find at first, follow that across that to Tottenham Crt Rd- then W to Marylebone High Street- then Paddington and then R up Eastbourne Terrace and then left and onto Westbounre Grove, then up to Westnourne Park Road and then down to Portobello Rd. You then should pop out near Ladbroke Grove Tube and then just head west down Cambridge Gardens( i think ) and you should hit Shepherds Bush. You can see the route in the above post ; which is bit far north for my liking.
Obviously, I can see the ride better than I can describe it, but dont go down the Bayswater Rd..

^ something like this: http://goo.gl/maps/4ix9 ?
I couldn't quite make sense of your directions, but that's an interesting route. I think I would still stay north of the Regent's Park though, if only because I currently commute on the Bloomsbury Cycle route and it's awful, full of crap cyclists, cars turning across it and pedestrians jumping in front of you without looking.

Yes, that's the route - but I went down Westbourne Park Rd ..
The Bloomsbury path is rather mad..

thanks for this guys! some real effort has gone in to these answers, appreciated. was expecting a couple of one-like answers, so this is great.
turns out i'm not gonna have to start this commute for a month or so, which gives me time to investigate a few of the options
Looks like I'm going to have to do this as a commute for a month or so starting Monday (only found out today, thanks work). I don't know West London at all, and was wondering if anyone has any route tips?
I've been thinking of going around the top of regent's park, through some of the back roads, and joinng Bayswater Rd around Queensway somewhere.
Ideally I'd like a route where I'm not going to have to stop at lights every 50m - is Bayswater Rd like that? I can imagine it being one of those shitkickers you get in London that's rammed with both traffic and red lights. Does it have a decent bus lane to cruise down?
