When my team were still replying on 5 year old servers running SQL Server 2000, SQL Server Profiler was a good tool for analysing live activities. We moved to a 2008R2 shared cluster about the same time as we got the ANTS performance suite, so I don't know how you analyse the queries that are hapening in versions since 2005 (When SSMS was introduced).
