Good bike fit, well selected contact points, tyre size'n'pressure......etc.
All of these things are going to have a bigger influence to comfort than frame material.
Get the Look, and build it up carefully. Fit a 25mm rear tyre, get a nice carbon seatpost, and bars that allow you to adjust the angle of your back.
