I am unsure of the benefits of padded, too, but I've never ridden much over 100-120k. I have only used padding on the last few, since January, and in general I'm happy with it. Always a bit of sudocreme for longer (50k+) rides.
But all in all I'd rather have airy bits than padding. I don't like the nappy bulky feeling at all. Think I'm giving it a miss for the Cambridge reccy tomorrow.
