100 miles on a fixed (especially that gear) is going to be a killer, especially if you haven't been training for it. there's a guy (sometimes on here, sometimes on birmingham cyclist) who does massive fixed rides, but has been building up over years to do super-audaxes (think that's what the crazy 500 mile plus rides are called anyway).
pacing yourself and not blowing all your energy is really hard on a fixed - i built up to around 100k last year, and i suppose i could go further if i really had to, but it won't be much fun, especially the second half. make sure you take loads of food and drink and eat often otherwise you'll blatantly bonk.
