I know bits of the route, and the bits I know are fairly flat, but it still seems to go up Devil's Dyke (which I haven't been up, but it's still rumoured to be a bit of a climb).
My favourite route to Brighton is the L2B Veteran Car Run route. That really is flat--it has to be, for 100+-year-old cars--, and only has two hills in it. It gets into Brighton via Clayton Hill, which is just north of Pyecombe on the map you linked to. It's not a very big hill at all. I can't remember where the other one was, much as I can't remember too much of the route, as it was all signposted, but quite a bit of it was on fairly major roads, including the lovely A23 for the final run into Brighton.
