i built a singlespeed with vertical drop-outs and used a chain tensioner to accomodate the lack of horizontal 'pull'. I love it but i dig that you might not be happy rolling freewheel. (you cannot use a tensioner if you want it to be fixed)
if you have your heart set on fixed then i'm told you can pull it off with vertical drops but your choice of gearing is very limited and still you might not be able to achieve sufficient tension in the chain. This calculator will help - http://eehouse.org/fixin/fixmeup.php
