Personally I'd be getting two bags for that.
Something weatherproof and comfortable for cycling and something more like the linked one with a clamshell (I've got an earlier model of this for work trips https://www.tortugabackpacks.com/products/expandable-backpack ) for work trips.
Getting something that does both will just be a compromise for one or the other.

The seams on my Chrome Urban Ex backpack have given up.
It's actually the third bag I have had through their lifetime warranty that has given out like this.
Not keen on another of the Chrome Industries heat welded seam bags, they just aren't up to it.
I do a cycle, train, cycle commute in all weather and do a fair amount of 2 or 3 day flight trips for work so looking for something that can handle both.
