I reckon so if it's on the same line. Last time I got the train from Eastbourne, I had to change at Brighton.

I’d expect it to be valid. Most flexible train tickets are valid over a wide range of alternative routes.
Use the Advanced option on the National Rail to route Via Brighton and if it still offers you the same ticket it is valid. Take a printout / screenshot if necessary.
