Weight! The problem with a lot of modern litre bikes is dry weight. They produce amazing power but they are not as light as a 250cc 2 stroke. The 2 Stroke runs out of performance earlier but the lighter package makes it lots of fun all the time you're cornering and racing between corners.
A great new era for m/bikes would be electric without the weight. Physics is a bitch though.
Other thing worth remembering is motorbiking is not only about dry days and track days. A 500cc bike is plenty fast enough screaming across the countryside in the middle of the night in winter when every element is trying to kill you.
