Does it matter that the route starts/ends in Winchester, but I will be starting out from Reading?

Garmin will just pick up the route. But it will "finish" when it gets to the routes finishing point and you'll have to restart it.

You can change the start/end points of a route in RWGPS too though so you could fix it in advance. I rarely bother though.
