Is this safe to do without a wheel-build? I have some 650b rims and want to test some tyres for size in my frame but can't afford the hubs and spokes atm. This would satisfy my curiosity.

Yeah, fine AFAIK. Even pressure all round. Just don't whack them up super high

What'll a wheel build do? Spokes would be pulling the rim in the same direction the tyre is pushing it.
Stretching tubs over an unbuilt rim is a common practice and you could be putting 200+ psi in them.
