Auchentoshan is just north of the airport. I'd hit that and then turn around. Or Glengoyne is a bit further but worth the trip...though perhaps not if you have to drive afterwards. Otherwise, I don't think there's anything of note south of Glasgow.

I can vouch for both the Auchentoshan and Glengoyne tours. Auchentoshan if you can only pick one.
The only distillery south (on the mainland) is Bladnoch and that's closer to Stranraer than Dumfries. It's not one I've tasted so can't help there.
