You could use the LOL Steam Ride route from London to Oxford - GPS lives here: http://www.aukweb.net/events/detail/14-599/
From there, if you get onto National Cycle Network route 57, that will take you to Northleach, where you'd need to devise your own way to nearby Cheltenham, which is about 13 miles. NCR 57 is supposed to go the whole way from Welwyn Garden City to near Gloucester but is sadly incomplete and is missing the section between Oxford and Witney, so you'd need to get creative between those two points too, though it's only 12 miles. Sustrans warns that some of the route is off-road, but I've had a good look at Witney to Northleach and it's all lanes. NCR 57: http://www.sustrans.org.uk/ncn/map/route/route-57

Here's my Lewisham to Whitstable route as ridden with @madeinmelburn .
Goes over the North Downs which is less direct but far more pleasant. Pilgrims Way and Hollingbourne make this a lovely ride. Tried and tested. -

This was nice. On the return route go east or Ardingley resevoir, west of weir wood though and avoid crawley.

bit of a long shot here, but has any one got any nice loop rides from south east london (nunhead) out into the lanes, probs north downs-ish, that would be ok on fixed. want to go for a ride with gf but our road bikes are in the shop, dont want anything to hilly! as i said, may be a long shot.. Ta!

The route of the ladies owl ride (and back) was north Kent but pretty flat, I'll dig it out when I get home.
